DNA 损伤会引起基因组不稳定,诱发损伤修复,激活一系列细胞信号通路决定细胞命运:修复,凋亡或衰老。基因组不稳定性也被认为神经退行性疾病,癌症和免疫炎症的重要特征之一。
本课题组聚焦DNA损伤修复和表观遗传调控之间的互作及其对基因表达和细胞命运的作用是一个新颖而又关键的问题。主要研究内容包括:
1. DNA损伤在神经系统分化发育、可塑性及神经退行性疾病中的作用机制
神经元存在着极高水平的DNA单链损伤(single strand breaks,SSBs)和大量的双链损伤(double strand breaks,DSBs),本课题组通过各种生化分子,基因编辑和课题组已开发的检测DNA断裂和修复过程的高通量测序技术等手段,解析DNA损伤修复在神经系统分化发育及神经退行性疾病中的机制和功能。
2. DNA损伤与炎症及记忆形成
研究炎症与记忆形成的分子机制对于深入理解细胞衰老和神经退行性疾病至关重要,近期的研究表明DNA损伤与表观遗传修饰改变可能是炎症及记忆形成的重要事件,因此阐明DNA损伤与表观遗传调控在上述过程中的功能机制是本课题组的重点关注方向。
3. 非编码RNA及其他表观遗传因子对DNA损伤修复的作用
DNA损伤修复通路会受到表观遗传的精细调控,从而维持基因组的稳定性和DNA复制的准确性,我们将利用多组学研究技术研究非编码RNA及其他表观遗传因子在DNA修复过程中的功能机制,寻找其中重要的新分子靶点。
